Psilocybin Test Kit Validation and Case Report
In this Case Report, we demonstrate the precision and reliability of the Psilocybin Test Kit for measuring psilocybin potency. Dried Psilocybe cubensis Albino AA+ fruiting bodies were first finely ground. Different amounts of this powdered biomass were accurately weighed and placed into separate Eppendorf tubes for extraction.
Each sample was individually extracted and filtered, then incubated with the test kit's detection solution in cuvettes. After incubation, the samples were measured using the QTest Photometer at 595 nm, providing accurate results based on the color change.
Simultaneously, three 50 mg samples were extracted and analyzed using HPLC to determine the exact psilocybin content, ensuring the accuracy of the results.
The resulting plot shows a highly linear correlation between the color change and psilocybin concentration, confirming the effectiveness of the Psilocybin QTest. The data exhibits a strong linear relationship (R² = 0.9984), validating the Psilocybin Test Kit as a reliable method for measuring psilocybin content.
This experiment highlights the precision and reliability of the Psilocybin Potency Test, demonstrating how the Psilocybin QTest provides an easy-to-use, scientifically backed solution for determining mushroom potency with confidence.

Absolutely! The PSILOCYBIN QTest is perfect for testing the potency of any psilocybin-containing mushrooms, including popular species like Psilocybe cubensis, truffles, and even mycelium. It requires only 0.15 g of dried material to perform the test.
No, the PSILOCYBIN QTest is specifically designed for use with dried fungal materials, such as mycelium, fruiting bodies, or truffles. Fresh mushrooms contain a high and fluctuating water content, which can interfere with accurate measurements in this psilocybin test.
Yes! In addition to the application on dried fungal material, the rapid test shows equally reliable results for most solvent-free extracts.
Extracts are usually more concentrated. To ensure that the color result of the PSILICYBIN QTEST still fits the scale on the evaluation card, you have to reduce the amount of starting material accordingly. Before testing homogenize your substance. Typically, methanol extracts are triple-concentrated, so if you have a crude extract, we recommend dividing the amount you use for the test by the factor three (50 mg). If the extract is considerably more concentrated, reduce the initial quantity accordingly. The extract must be as solvent-free as possible, as this can interfere with the extraction solution.
Remember that you have to multiply the result of the color scale in the end with the used factor to get the actual result for the remaining sample. For example, weigh 50 mg (0.05 g) of material for a 3X extract. Weigh 15 mg (0.015 g) of material for a 10X extract.
No, the PSILOCYBIN QTest is specifically designed to measure psilocybin and psilocin content in magic mushrooms and does not work with synthetic substances like 4-AcO-DMT.
The PSILOCYBIN QTest quantifies the total alkaloid content (psilocybin + psilocin) and gives the added values in the form of a total concentration of psilocybin.
The background for this is that only the very stable active ingredient psilocybin is naturally present in healthy fungal tissue. For example, if not properly dried and stored, degradation of the prodrug psilocybin to unstable psilocin, the actual pharmacological active ingredient, occurs. Even if this occurs, the PSILOCYBIN QTest will give you information about the total amount of both alkaloids. Both substances have different molecular weights and to avoid a complicated conversion we give the value in terms of the well-known alkaloid psilocybin.
The PSILOCYBIN-QTEST works well with, for example, rotated or evaporated ("dry") extracts, but also some solvents like ethanol work if the concentration of alkaloids is not too low. This means that if too many foreign solvents are added to our extraction, it will disturb our extraction system.
The best way would be to evaporate the ethanol or any other solvent from a defined amount, note the dry weight, and measure it with the test.
The PSILOCYBIN QTest has a shelf life of at least 18 months if stored properly. Keep the reagents in the refrigerator at (35 - 46 °F/ 2 - 8 °C) and out of direct sunlight. Over time, the extraction liquid may turn slightly yellow, that’s okay. It is still usable.
Yes we have a validation for our PSILOCYBIN-QTests. This is a voluntarily validated and standardized test procedure based on the pharmaceutical method validation guideline ICH Q2(R1), which we have extended to the QTests. This guideline describes the requirements of the analytical method for active ingredients that are subsequently to be used in human medicine and guarantees a safe dosage of these active ingredients, for example in pharmacies.

We recommend ensuring that floating fungi are not sucked into the system and positioning the sterile filter below the floating fungus layer before applying vacuum. If the process fails initially, cleaning the filter very carefully and reapplying the syringe can help. The test can still function if fungal components are transferred, but this may complicate color evaluation. Link to the video.

Dr. Felix Blei is the CEO and Founder of miraculix Lab, and a Postdoctoral Researcher at the Friedrich Schiller University Jena. With a PhD (Dr. rer. nat.) in Microbiology and a strong background in natural product biosynthesis, he is internationally recognized for his pioneering work on psychoactive fungi—particularly the biosynthesis of psilocybin and related compounds (Blei, F., 2020).
During his doctoral research at the Institute of Pharmaceutical Microbiology under Prof. Dirk Hoffmeister, Dr. Blei was the first to elucidate the full biosynthetic pathway of psilocybin in Psilocybe mushrooms (Fricke, Blei et al., 2017) Angewandte Chemie. He further developed an in vitro system capable of producing psilocybin, serotonin, and novel non-natural analogues (Blei et al., 2018) Chemistry- A European Journal . His discovery of naturally occurring β-carbolines in Psilocybe—compounds that may synergistically enhance psilocybin's neurotropic effects—led to the concept of “psilohuasca,” a naturally occurring combination of MAO inhibitors and psychedelics in fungi (Blei, Dörner et al. 2020) Chemistry–A European Journal.
Building on his academic expertise, Dr. Blei developed the first reliable rapid tests for psilocybin, which laid the foundation for the spin-off company miraculix. Today, miraculix provides quantitative drug checking tools that are used throughout Germany and Europe. He also leads the German ALIVE project—an evidence-based drug checking Initiative funded by state governments, offering mobile substance analysis and harm reduction at music festivals and public events.
